Yes, on the grounds that Saitama gets dunked on by a mosquito and an alley cat because it's funny. Therefore, Bugs dunking on him would also be funny.

Going off of their usual MO? Bugs would try to trick and annoy the heck out of Saitama, and Saitama doesn't take the bait.
Drop and anvil on Saitama, it just bounces off his head and Saitama says that anvils are dangerous. Try and trick Saitama with fake signs and Saitama just never even bothers to read the sign at all, or reads it and avoids the trick because he is focused on something inane.
Saitama has very flat reactions to things because he is emotionally dead inside. Bugs won't get the satisfaction of making Saitama mad.

It's not about whose win would be the funniest but rather who is funnier in defeat. In which case I would have to give it to Bugs, who historically has experienced more defeats that have been comedic.

Saitama is basically one short step from becoming a completely hollow meat-robot. That, or ascending into a Buddha.
Saitama does feel annoyance, but it's to impossibly mundane things, like a mosquito that keeps evading his swats, or losing rock-paper-scissors a series of times in a row. Grandiose antics like dropping pianos on him don't bug him, but if he can't get to the store in time for the big sale on meat, God help you if you're the cause.

Not really the best example, since that short was Bugs playing the famous role of a classic fable character who is always destined to lose. If Bugs is presented as the protagonist in the story, then he is likely going to win or at least achieve some kind of satisfaction over whoever he is harassing. Granted, the most he could probably do with Saitama is just annoy him to the point that Saitama either gets mad and leaves in a huff, or finally realizes he has other business to attend, like maybe getting to a sale.

It solely depends on who starts the conflict. The antagonizer would be the loser.
Pretty sure Bugs had suffered some humiliation even when he's not the once to start aggressions, he just didn't pay nearly as bad as the offending party.
Frustrate like the mosquito and survive yes, beat him in a direct fistfight (which he would never do mind you because that's not funny) no.

If he's trying to catch, it, probably not, since the point of the Roadrunner is that he's never meant to be caught. If the two are just racing each other, I'd imagine Saitama would win since he can achieve incredible speeds if motivated enough, kind of like how Freakazoid can speed around the globe if something motivates him to, yet he rarely did and thus didn't use his full speed for most of the show.
Saitama is fast, but he's not that fast. He's spec'd into Strength. It would be the mosquito situation again.
Literally traveled to the moon in an instant. He's as strong and fast as the plot demands.
