Comedian was the original and real Joker. Bruce confirms it when he admits that he knew the Joker's name one week after they first met and the Killing Joke flashbacks just back it up. Everyone saying the Criminal just wants him to be it.

That scene just means he knew who the only Joker around by that point was. I don't think it means much regarding the others one way or another since they aren't even mentioned in that scene and are dead by that point.

Note that previously the other Jokers had even called Comedian "a failed comedian", so it's not like they were claiming/believing they had his backstory.

Batman outright says the Criminal is the one he encountered fist. The other Jokers came later.

The ending states Killing Joke is legitimately the origin of the Joker.
The wife is alive and so is the kid.
Batman figured who the Joker was a week after he appeared.
This is literally the ending.
