It's pretty clear by the Sasuke flashback at the Valley of the End that Itachi was intended to be a double agent in the first place.

But why does he tell Sasuke to kill his best friend ?

Itachi was the "perfect" shinobi but that's not a good thing to be either. He was a peaceful person at heart who was constantly at war with himself and believed that you had to make hard decisions in life no matter what. He was killing himself doing the things he thought he had to do, but the very shinobi world he lived in was one that was creating these awful things he had to choose between. Naruto and Sasuke both idolize him as someone who always did the best he could but Itachi was never somebody in control of his life and often made poor decisions because of it.

He didn't. He simply explained to Sasuke that to obtain the Mangekyou Sharingan, an Uchiha had to kill their best friend. This turns out to be a half-truth because it's not the act or murder but the pain of loss that triggers it, which tracks with the Sharingan being something that is unlocked when an Uchiha reacts very strongly to the need to protect someone/the need to avenge the loss of a loved one. My theory here is that he was intentionally trying to make Sasuke hate him, so he told him that in order to obtain the same power as him he would have to do something truly repugnant in the hopes that Sasuke would reject the power (which he ultimately does).
