Realistically I don't see a U6-only spinoff happen. If the girls return, it will be when the conflict reaches a multiversal level in the final arc or so, where every universe has to fight
If we're talking pure headcanon I'd really love a Sadala arc showing us some Caulifla, Kale and Cabba adventures. A lot could be done with them, for example exploring their parallels to Goku, Broly and Vegeta

It's a sad fact that anime/manga have to make bank. That's why the Broly movie was so lowest-common-denominator, it needed mass market appeal. And the numbers don't lie. So yeah, as amazing at it would unquestionably be I don't foresee any kind of solo appearance either.
I'd like to see those as well. They could be fun, maybe hark back to the OG Dragon Ball days.

I don't know how well-received the U6 parts were in the anime for me to judge how much expectation there is for Toyo to do something with them again. How he included them in the ToP was definitely underwhelming though.

It was, my main complaint is basically Kale being such a non-character in the manga. Can't say how the anime did her relation to Caulifla was perfect but it's better than those two barely even knowing each other
Also the Berserk form, anime Kale gets to master and make it her own while MANGAle stays mindless and helpless until the fusion forces her back to sense

Given her depiction and dialogue snippets from the anime, it was pretty obvious that people could infer some deeper bond or connection between them. But, this being Dragon Ball, no hint of romantic interest would ever be depicted. Hell, even the longest-standing women from Dragon Ball have little to no romantic dimensions to their character.
As for her forms, they were basically two sides of the same coin. Different interpretations for different audiences. They both had positives and negatives.
