It's funny because Mario, the titular character of the video games series Super Mario becomes bigger and stronger when he picks up a mushroom-shaped item. Here you can see that after eating a pizza with mushrooms, Mario has become so big he broke his chair and can't fit inside the room anymore. The waitress is surprised because that's an usual thing. The joke is that you don't usually think of the mushroom item interaction outside of a video game setting. Here you see how inconvenient it would be if eating mushrooms made you bigger in real life. The fact that Mario is of italian descent and is eating a pizza adds another layer to the joke.
