They're supposed to be mutants so them looking different is fine. I think in the comics or somewhere it was revealed they're not even technically brothers they were just all put together at the pet shop. I think that works because it shows none of that matters because they're still family.

It's something they teach in art schools apparently. The strong/tough character has to have a big and bulky silhouette so that you know that they are the strong/tough one. They did the same thing with Knuckles in Sonic Boom.

Implying every brother looks exactly the same. They are brothers, not twins. Besides, the look is different because they are different type of turtles. Its not like they are literally brothers, they are turtles the bad guy grabbed from somewhere and mutated with Splinters DNA.
